چکیده

The copper(1) complex [(TMPA)Cu(RCN)]+ (1) binds 0 2 forming [{ (TMPA)Cu}2(02)]2+ (2), with trans-y-1, 2 peroxo-coordination. Ligands with quinolyl groups substituting for the pyridyl donors in TMPA cause dramatic changes in the course of reaction, in one case stabilizing a CdO2 1:l adduct [(BQPA)Cu(02)]+ (6 ) . The kinetics/thermodynamics are compared. Reaction of 1 with (F 8-TPP)Fe(II)pip2 (8) and 0 2 yields the p-0x0 species [(Fg-TPP)Fem-O-Cun(TMPA)]+ (9); this reversibly protonates giving p-hydroxo bridged [(Fg-TPP)Fe-(OH)-Cu(TMPA)]2+ (12). The novel NMR properties of 9 are described. These complexes are discussed in terms of their model 02-chemistry in hemocyanins or cytochrome c oxidase.

Role of copper ion in regulating ligand binding in a myoglobin-based cytochrome C oxidase model.

Cytochrome c oxidase (CcO), the terminal enzyme in the mitochondrial respiratory chain, catalyzes the four-electron reduction of dioxygen to water in a binuclear center comprised of a high-spin heme (heme a(3)) and a copper atom (Cu(B)) coordinated by three histidine residues. As a minimum model for CcO, a mutant of sperm whale myoglobin, named Cu(B)Mb, has been engineered, in which a copper at...

Intracytoplasmic Copper Homeostasis Controls Cytochrome c Oxidase Production

UNLABELLED Copper is an essential micronutrient used as a metal cofactor by a variety of enzymes, including cytochrome c oxidase (Cox). In all organisms from bacteria to humans, cellular availability and insertion of copper into target proteins are tightly controlled due to its toxicity. The major subunit of Cox contains a copper atom that is required for its catalytic activity. Previously, we ...

Probing protonation/deprotonation of tyrosine residues in cytochrome ba3 oxidase from Thermus thermophilus by time-resolved step-scan Fourier transform infrared spectroscopy.

Elucidating the properties of the heme Fe-Cu(B) binuclear center and the dynamics of the protein response in cytochrome c oxidase is crucial to understanding not only the dioxygen activation and bond cleavage by the enzyme but also the events related to the release of the produced water molecules. The time-resolved step-scan FTIR difference spectra show the ν(7a)(CO) of the protonated form of T...

X-ray absorption edge studies on oxidized and reduced cytochrome c oxidase.

The x-ray absorption edge spectra of the Cu and Fe-centers in oxidized and reduced cytochrome c oxidase (ferrocytochrome c:oxygen oxidoreductase: EC 1.9.3.1) have been obtained using synchrotron radiation from the SPEAR storage ring at the Stanford Linear Accelerator Center. In addition, oxidized and reduced plastocyanin as well as a number of model copper compounds in various oxidation states ...
